Discord recommends custom emoji at 128 × 128 pixels in a square (1:1) aspect ratio. Importantly, the old rule that emoji had to be exactly 128 × 128 in PNG only is now outdated — Discord's uploader automatically resizes and crops any image you give it. You can upload a larger square (256 × 256 works well) and Discord will scale it down cleanly.
The upload size is not the display size. In chat messages, emoji render at roughly 32 × 32 pixels, and in reactions at about 48 × 48 pixels. An emoji sent on its own in a message (a "jumbo" emoji) renders larger. Either way, your design needs to be readable at thumbnail scale, not just at the 128px source.
